Global Clinical Landscape & Future Outlook
WAC is a critical intracellular adapter protein characterized by its WW and coiled-coil domains. It plays a vital role in epigenetic regulation by interacting with the RNF20/RNF40 complex to promote histone H2B monoubiquitination, and it independently functions in transcription and autophagy pathways. Mutations in the WAC gene are the primary cause of DeSanto-Shinawi syndrome (DESSH), a rare neurodevelopmental disorder.
Currently, WAC is an exploratory/preclinical target. There are no active clinical-stage therapeutics (such as small molecules or gene therapies) directly targeting WAC. However, the global R&D landscape is actively investigating WAC's role in neurodevelopment and oncology. Future translational efforts are expected to focus on conceptual gene replacement strategies for DESSH, as well as exploring the WAC-RNF20/40 protein-protein interaction (PPI) interface as a potential vulnerability in specific cancers.
